CRISPR/Cas9 and Base Editing, principles and applications

Niveau
Introduction
The aim of the course is to provide participants with insight into the principle of CRISPR/Cas9 gene-editing methodology and gene-editing without the formation of double strand breaks via Base Editing. A thorough insight is given into the many different applications of the techniques as academic gene modification tools as well as the derived translational aspects in biotechnological and therapeutic contexts. Regarding the latter, examples of FDA approved Cas9 based gene therapies will be given, along with ethical considerations regarding use of curative Cas9 based gene therapies of severe diseases. The course is primarily aimed at laboratory technicians and bioanalysts, or others with a desire for in-depth theoretical insight into CRISPR can also participate with advantage
Participant requirements:
Experience with standard molecular biology, DNA and cell culture techniques is an advantage but not a prerequisite.
Contents:
- Review of gene targeting before and after the “CRISPR craze” era in 2013.
- Brief review of the principle of gene editing based on alternative methodologies such as zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs) and Meganucleases.
- Detailed review of the principle of using CRISPR/Cas9 as a gene editing technique, with particular focus on its use in eukaryotic cells.
- Review of methods for the identification of Cas9 induced genome changes.
- Review of derivative CRISPR/Cas9 applications with special emphasis on Base Editing.
- Review of CRISPR specificity and safety, including the unintentional “on target” and “off target” concept. • Review of the biotechnological and therapeutic application possibilities for CRISPR/Cas9 and Base Editing, with particular focus on CRIPSR/Cas9 and Base Editing based gene therapy.
- Review of FDA approved Cas9 based gene therapies.
- Ethical reflection on the use of gene therapies.
- Practical case-based exercises regarding gRNA design for your own project, ethical considerations for CRISPR, etc.
The organization of the teaching:
Theory teaching alternating with cases and assignments.
